Evosep High-Throughput Proteomics Separation System
Liquid-mass spectrometry (LMS) technology is widely used in proteomics. However, the current liquid-liquid mass spectrometry (LLMS) technology has the problem that the speed of liquid-phase separation is difficult to match the speed of mass spectrometry analysis, which reduces the working efficiency in proteomics research, and the Evosep one can be equipped with mass spectrometry such as Thermo Q Exactive HF-X/HF, Bruker TimsTOF Pro, and Sciex Triple TOF 6600, which can greatly shorten the sample analysis time and meet the demand of high-throughput proteomics research.
Evosep One Principle
The Evosep One is an Evotip-centered instrument for high-throughput proteomics applications with high-resolution mass spectrometry.The Evotip is a disposable capture column in the form of a pipette tip, which allows for desalting and decontamination of samples in the Evotip.There are five system pumps in the Evosep One, including four low-pressure pumps and one high-pressure pump. The low-pressure pump is used to elute samples and gradients from presets in the Evotip. The high pressure pump is used to push the liquid in the storage ring into the analytical column connected to the mass spectrometer for high throughput proteomics sample separation.

A fast and highly robust analytical method based on the Evosep + Sciex Triple TOF 6600 was successfully developed and used for the study of mouse liver tissue, with 460 injections in 8 days at a gradient time of only 21 min/sample, as opposed to the conventional method with a gradient time of 90 min/sample. As can be seen in the figure below, the retention time was reliably reproducible and the data were highly consistent.
Evosep Application Case II
The researcher successfully developed a PRM assay for the detection of doping rHuEPOs (recombinant human erythrocyte proteins) in racehorse plasma based on Evosep + Thermo Q-Exactive HF, which requires only 11.5 min gradient time/sample to complete the analysis of one sample. As can be seen in the figure below, the researchers first validated the method by spiking blank plasma samples with alfa Darbepoetin and obtained highly reproducible retention times for its characteristic peptides T6 and T17 at different spiked concentrations, respectively. Afterwards, the researchers applied this method to the sample assay and obtained the concentration of rHuEPOs in the plasma samples from racehorses.
